Indole-3-acetamide (IAM) is an intermediate of the indole-3-acetic acid (IAA) biosynthetic pathways in plants. It is known that the combination of IAA and horseradish peroxidase (HRP) induces apoptosis in cancer cells. Here, we investigated effects of IAM/HRP treatment on PC-3 human prostate cancer cells and TCCSUP human urinary bladder carcinoma cells. First of all, we evaluated the effect of IAM on PC-3 and TCCSUP cell viability. IAM had no cytotoxic effects on PC-3 and TCCSUP cell viability. Next, we examined whether IAM/HRP could produce free radicals like IAA/HRP. DCF assay demonstrated that IAM/HRP produced free radicals in a dose-dependent manner, which is suppressed by Trolox, a water-soluble vitamin E derivative. Although mechanisms of IAM/HRP-induced free radical production remain unknown, we examined whether IAM/HRP could affect the proliferation of PC-3 and TCCSUP cells. MTT assay revealed that IAM/HRP decreased the proliferation of PC-3 and TCCSUP cells. Therefore, we suggest that IAM/HRP may provide a new potential candidate for urinary organ cancer treatment.
