Assessment of biological activity in vitro and phytochemical constituents from Rosa hybrida leaves

The health benefits of roses have been demonstrated previously. This study aimed to isolate phytochemical constituents from the leaves of Rosa hybrida (LRH) and identify their chemical structures by1H-NMR,13C-NMR, and mass spectrometry. Fractionation was carried out using open-column chromatography to isolate the compounds in n-hexane, chloroform, ethyl acetate, and n-butanol. Four phytochemical constituents from LRH were isolated and identified, including oleanolic acid, uncaric acid, quercitrin (quercetin 3-O-rhamnoside), and afzelin (kaempferol 3-O-rhamnoside). Furthermore, the study explored the antioxidant properties of the extract and assessed the total contents of polyphenols and flavonoids. The LRH extract exhibited strong antioxidant activity, with IC50 values that were on par with the positive control used in DPPH and ABTS assays. This study highlights the potential medicinal value of LRH. Future studies should focus on the applications of the isolated compounds in health and conventional medicine or as natural antioxidants in the pharmaceutical and food sectors. Further studies may be required to investigate sustainable cultivation and harvesting of LRH for its phytochemical components. © The Korean Society for Applied Biological Chemistry 2024.
