Dual-mode chlorogenic acid detection via competitive Boronate displacement: Smartphone-assisted monitoring during coffee roasting

Chlorogenic acid (CGA), the predominant polyphenol in coffee beans, is essential for antioxidant activity and flavor. This work presents a dual-mode sensing platform combining red-emissive B, N-doped carbon dots with 3-nitrophenylboronic acid-bromopyrogallol red complex for rapid CGA quantification. The mechanism exploits competitive displacement where CGA's catechol moiety binds 3-NPBA, liberating bromopyrogallol red and producing colorimetric response with fluorescence restoration via inner filter effect relief. The fluorometric mode achieved 1.0–190.0 ng/mL linearity (Limit of detection: 0.29 ng/mL) while colorimetric mode spanned 0.5–4.5 μg/mL (Limit of detection: 0.18 μg/mL), with excellent recoveries of 96.0–102.9% and 98.8–100.5%, respectively. Smartphone integration enabled portable detection with linear ranges of 10.0–110.0 ng/mL (fluorometric) and 1.0–17.0 μg/mL (colorimetric). Application to Arabica beans revealed progressive CGA degradation from 120.25 mg/g (green) to 13.41 mg/g (dark roast), demonstrating monitoring for coffee quality control and roasting optimization.
