This study compared mesenchymal stem cells (MSCs) isolated from the perirenal (P-AMSCs) and subcutaneous (S-AMSCs) adipose tissues of three 30-month-old Hanwoo cattle (n = 3), with both tissues collected from each animal. Flow cytometry showed both expressed CD29/CD73 and lacked CD34/CD45, but CD105 was higher in P-AMSCs (26.3 %) than S-AMSCs (1.2 %). P-AMSCs exhibited faster proliferation, shorter doubling times, and higher cell counts and MTS assay results (p < 0.0001). Notably, during proliferation, S-AMSCs exhibited significantly higher FABP4 and FASN expression than P-AMSCs (p < 0.05). Adipogenic differentiation revealed greater lipid accumulation and higher expression of PPARγ, FABP4, LPL, and FASN in P-AMSCs (p < 0.001), with Oil Red O staining showing higher differentiation (10.95 % vs. 7.26 %, p < 0.05). P-AMSCs also showed stronger osteogenesis (91.8 % vs. 60.5 % mineralization, p < 0.0001) and upregulation of COL1A1, RUNX2, DLX5 (p < 0.001), and enhanced chondrogenesis with increased SOX9, COL2A1, and ACAN (p < 0.01). These findings indicate P-AMSCs have superior proliferation and differentiation potential, supporting their use in adipose tissue engineering and cultured fat applications.
