Concentration-Dependent Effects of Arbuscular Mycorrhizal Fungi on Soil Mycobiome Under Individual Pot-Based Greenhouse Cultivation

Arbuscular mycorrhizal fungi (AMF) are well-characterized plant symbionts that form mutualistic associations with host plant roots, contributing to nutrient and water-use efficiency and enhancing tolerance to external stresses. To reduce environmental variability, this study evaluated the concentration-related effects of AMF supplementation on soil mycobiome dynamics under individually potted, environmentally controlled greenhouse conditions. Strawberry (Fragaria×ananassa cv. Puget Crimson), a crop influenced by soil and rhizosphere conditions, was used as the host plant. A commercial AMF inoculant, MycoApply® Injector Endo, was applied biweekly at two concentrations, namely, T1 (1.25%) and T2 (2.5%), alongside a water-only control. Soil fungal communities were assessed in baseline soil collected before AMF treatment and in control, T1, and T2 soils collected after 7 weeks of cultivation, followed by internal transcribed spacer amplicon sequencing and analysis using the QIIME2/DADA2 pipeline. Alpha-diversity indices showed no significant differences among treatment groups, whereas beta-diversity analysis revealed significant differences in fungal community composition (permutational multivariate analysis of variance, PERMANOVA: R2=0.105, P=0.001). Venn diagram analysis further indicated that T2 harbored the highest number (745) of unique amplicon sequence variants (ASVs), suggesting that the higher AMF concentration was associated with the largest number of treatment-specific ASVs. Linear discriminant analysis effect size revealed enrichment of taxa previously associated with saprotrophic activity and biocontrol potential, including Sordariales and Fusicolla, in T2, whereas T1 was characterized by taxa reported to include rhizosphere-associated or plant-beneficial members, such as Podospora and Sebacinales. In contrast, the control soil was characterized by taxa that included species previously linked to opportunistic pathogenicity, while the baseline soil contained taxa associated with untreated initial soil conditions. These findings suggest that AMF supplementation under controlled greenhouse conditions is associated with concentration-related shifts in the soil mycobiome, including changes in taxa previously linked to beneficial ecological functions.
