A Study on the Hepatic Granulocytopoiesis in Human Fetal Liver

초록

The ultrastructure and distribution of eosinophilic and neutrophilic granulocytopoietic cells in 36 human fetal livers from 6 to 40 weeks of gestation were studied by immunohistochemistry using antibody to common leukocyte antigen(CLA) and electron microscopy. 1. The CLA reactive hemopoietic cells appeared as a single cell for the first time in hepatic laminae and mesenchymal tissue in 7 and 8 weeks of human embryonic livers. These cells were more numerous at portal connective tissue and hepatic laminae with age, and were forming cell masses in portal areas after the 6 month of gestation. 2. The more active granulocytopoiesis of neutrophils and eosinophils in various developing states was present during 26 to 34 weeks of gestation. The eosinophilic granulocytopoiesis was more active than the neutrophilic granulocytopoiesis. 3. Granular endoplasmic reticulum increased in size and amount at promyelocytes and myelocytes, and decreased at metamyelocytes with increasing specific granules. Many large mitochondria of myeloblasts became less and smaller in metamyelocytes. The progressive clumping of chromatin and the disappearance of nucleoli occurred in the course of maturation. The changes of intracelluar organelles were similar in both the eosinophilic and the neutrophilic granulocytopoiesis. 4. The microgranules, granular vacuoles and immature eosinophilic granules progressed the various intermediate stages of maturation and finally transformed into mature eosinophilic granules. Two types of neutrophilic granules were formed by transformation from microgranules of myeloblasts and from various intermediate granules of promyelocytes.
