Development of an unroasted coffee powder certified reference material for the accurate determination of aflatoxin B1

An unroasted coffee powder certified reference material (CRM), designated as KRISS CRM 108–10-026, was developed to ensure reliable and accurate analysis of aflatoxins (AFs). The CRM was prepared by mixing Aspergillus parasiticus-inoculated raw coffee beans with blank raw coffee beans to achieve representative levels of target AFs. The material was freeze dried, milled, sieved, and homogenized. The final produced CRM was aliquoted into ∼15 g portions per unit and stored at −70 °C. Certification of the CRM was performed using an isotope dilution–ultrahigh-performance liquid chromatography–tandem mass spectrometry (ID–UHPLC–MS/MS) method. The CRM demonstrated excellent homogeneity, with relative standard deviations of 0.6 % for AFB1, 1.2 % for AFB2, and 1.0 % for AFG1. Stability studies indicated that the CRM remained stable under various storage conditions: −70 °C for up to 12 months. A certified value of 5.6 ± 0.1 μg/ kg was assigned for AFB1, while indicative values of 0.082 ± 0.003 and 0.67 ± 0.03 μg/ kg were assigned for AFB2 and AFG1, respectively, due to the lack of their traceability to the SI units. This CRM is expected to serve as a valuable tool for ensuring measurement accuracy and comparability in the AF analysis of coffee.
