Dexamethasone 투여에 의한 마우스 간에서의 항산화 효소들의 활성 변화

초록
Dexamethasone, a widely used agent for septic shock therapy and immunosuppressoin, was injected intraperitoneally to mice three times at doses of 2.5 mg/kg, 5 mg/kg and 10 mg/kg at 6 hour intervals and reactive oxygen species and antioxidant enzymes in liver were measured. Dexamethasone injections significantly reduced superoxide radical (p<0.01) and xanthine oxidase activity (p<0.05 at 10mg/kg). A cytosolic antioxidant enzyme, Cu,Zn-SOD activity was significantly increased by dexamethasone treatment, whereas dexamethasone treatment reduced activity of a mitochondrial antioxidant enzyme, Mn-SOD. Glutathione peroxidase and catalase activities were decreased by dexamethasone treatment. Catalase protein level was significantly reduced by dexamethasone treatment, while it had no effect on glutathione peroxidase protein level. When normal liver was treated with dexamethasone, catalase activity was reduced in a dose-dependent manner, whereas its had no effect on glutathione peroxidase. Thus, these effects of dexamethasone treatment on various antioxidant enzyme activities in liver suggested that this anti-inflammatory drug could modulate oxidative stress under a variety of inflammatory conditions.
