An Immunohistochemical Study on the Dynamics of Alpha-Fetoprotein in HumanLiver During Normal Ontogeny

초록

The dynamics of alpha-fetoprotein (AFP) containing hepatocytes in 46 human embryos and fetuses ranging from 5 to 36 weeks of gestation were investigated using immunohistochemistry(monoclonal antibody, anti-human AFP rat IgG. The hepatocytes of 5 week embryo had the immunoreactive product against AFP with the granular pattern. About 30-50% of hepatocytes in the embryonic period showed the positive immunostaining for AFP with a perinuclear localization. The AFP containing hepatocytes had the tendency of the subcapsular distribution and with age, approximately 80% of hepatocytes were positive to the immunostaining. The staining pattern was changing from granular to diffuse at 4 months. The AFP containing hepatocytes were distributed throughout the parenchyme and frequently around the central vein and the portal area. The immunoreactivity of AFP containing hepatocytes became to decrease at the 8 months. The incidence of AFP containing hepatocytes decreased rapidly at 9 months and about 5% of hepatocytes showed the weak immunoreactivity in 36 weeks of gestation.
