The Structural Change of Rat Cerebral Cortex by the In Situ Perfusion withL-Glutamate, Glycine and Kynurenate

Rat brains were treated with in situ perfusion to investigate the presence and in vivo role of Gly2 receptor different from Glyl receptor. The change in cerebral cortex perfused with glutamate was similar to those receiving direct stereotaxical injection of glutamate, exhibiting axon-sparing effect. The swelling of dendrites in cortical layers 1 and 2 was more prominent than those in the other layers. The changes in cerebral cortex pefused with glycine was similar to those with glutamate, exhibiting swelling of dendrites especially severe in cortical layers 1 and 2. The result apparently represents excitotoxic reaction in cerebral cortex while glycine receptor in brainstem and spinal cord is inhibitory site. The weak excitotoxic reaction was discernible in the other cortical layers. The glutamate administration combined with glycine did not cause synergistic excitotoxic effect. Kynurenate, competitive glycine antagonist, did not exhibit apparent protective effect. So it is suggested that the glycine receptor in cerebral cortex especially in the cortical layer 1 and 2, exhibiting excitotoxic reaction to sufficiently high glycine concentration, is an excitatory glycine binding site.
