The investigation for prognostic markers are still evolving in nonsmall cell lung cancer inspite of marked improvements with target therapy and immunotherapy. Recently, LGI3(Leucine-rich-repeat LGI family member-3) was suggested as a prognostic marker in Non-small Cell Lung Cancer (NSCLC). This study was performed to define how the LGI3 was expressed among the different lung cancer cell lines with the conventional and real time RT-PCR, ELISA and Western blottings, which were analysed to relate TNF-α, EGF and cellular proliferation. Notably, NCI cell lines showed relatively higher expression in contrast to Korean SNU cell lines. LGI3 was released from A549 cells into the culture media. LGI3 dose-dependantly suppressed the proliferation of A549 cells. The LGI3 downregulated p21 protein expression and upregulated caspase-3 without change of p53. Taken together, we could find the different expression of LGI3 in NSCLC cell lines, which might explain the heterogeneity in its biologic behaviors and subsequent prognosis. With this findings, further studies may be needed to delve into the more exact roles in NSCLC.
