This study aimed to improve and validate the specification analysis method for copper sulfate, a fortifying nutrient used in food additives in South Korea. The existing method prescribed by the Korean Food Additives Code lacks clear conditions and procedures for saturating hydrogen sulfide to detect alkali and alkaline earth metals. A simple hydrogen sulfide generation device was developed to address these limitations, and the optimal reaction parameters were established, including diluted hydrochloric acid-to-ferrous sulfide ratios and reaction times. Under the optimized conditions, three replicate experiments were conducted, resulting in a residual weight of 3.20±0.30 mg, which satisfied the Korean Food Additives Code requirement of less than 4 mg and the Certificate of Analysis (CoA) specification of less than 0.03%. In addition, intra-day and inter-day analyses were performed to verify the reproducibility and optimization of the proposed method, yielding values of 3.04±0.28 mg and 2.78±0.51 mg, respectively. These findings confirmed the validity of the optimized method. This study is expected to enhance the safety of experimenters working with toxic gases and the general applicability of the analysis by providing clear conditions and procedures for generating hydrogen sulfide in detecting alkali and alkaline earth metals during the purity analysis of copper sulfate.
