Differential alteration of primary and secondary metabolites in chilling-stress related tissues of cold-stored Asian pears

Alterations in primary and secondary metabolites influence fruit quality in Asian pears, particularly under cold-storage stress, and these changes are cultivar dependent. In this study, we compared metabolic alterations among tissues affected by cold-induced disorders in healthy (unaffected flesh tissue), sound (asymptomatic tissue adjacent to browned flesh), and browned (symptomatic, severely affected flesh tissue) fruits of two Asian pear cultivars, ‘Wonhwang’ and ‘Chuhwangbae’, after cold storage. Primary metabolites, including soluble carbohydrates, organic acids, and amino acids, were significantly higher in the browned tissue of the ‘Chuhwangbae’ cultivar than in other tissue types. Meanwhile, the ‘Wonhwang’ cultivar showed higher mineral contents and volatile organic compounds (VOCs) in healthy tissues, followed by sound tissues. Moreover, primary metabolites associated with major pathways, such as glycolysis and the tricarboxylate cycle, were upregulated in the browned tissues of the ‘Chuhwangbae’ cultivar. Overall, these findings highlight the primary metabolic pathways and VOC profiles among tissue types, particularly, in cultivar-dependent susceptibility to cold-induced browning in Asian pears.
